چکیده

The effects of an extracellular sodium (Na) concentration of 200 mM on function, ionic exchange, and water movements were examined in the vascularly perfused rabbit intraventricular septum. When the perfusate was switched from 142 mMNa to 200 mMNa, dP/ dt fell 20.4% over 2 minutes; dP/dt then recovered to the control level. The time course of the functional recovery correlated with the uptake of Na into the slowly exchangeable fraction of the tissue, and the rate of recovery in 200 mM Na depended on the frequency of contraction of the preparation. During perfusion with 200 mM Na, a 41% increase in slow-phase Na content compared with the content during perfusion with 142 mM Na was observed. The hyperosmolar perfusate induced a 21% cellular dehydration within 10 minutes as estimated from measurement of 10-minute SO4 space and total tissue water. The time course of dehydration was much more rapid than the time course of functional recovery. Calcium (Ca) influx during the period when dP/dt had returned to control levels in 200 mM Na was the same as it was during a control period in 142 mM Na. Thus, there appears to be a Na-Ca exchange system which enhances Ca influx and thus causes an inotropic effect in response to an increase in intracellular Na concentration.

Calcium-dependent enhancement of myocardial diastolic tone and energy utilization dissociates systolic work and oxygen consumption during low sodium perfusion.

The relationships and correlations among functional, metabolic, and ionic consequences of low sodium perfusion were studied in isovolumic, retrograde-aortic perfused working rat hearts by 31P nuclear magnetic resonance, oxygen consumption, and atomic absorption spectrometry. Reduction of perfusate sodium from 144 to 74, 51, 39, and 25 mM in four separate groups of hearts via lithium substitutio...

A phosphorus-31 nuclear magnetic resonance study of the metabolic, contractile, and ionic consequences of induced calcium alterations in the isovolumic rat heart.

Isolated adult rat hearts perfused in an isovolumic mode were used to study the effects of sodium-potassium pump inhibition and sodium-calcium exchange alterations on the tissue content of adenosine triphosphate, phosphocreatine, inorganic phosphate, and intracellular pH, all measured by phosphorus-31 nuclear magnetic resonance spectroscopy. Rates of oxygen consumption, contractile function, an...

Ultrastructural Alterations Produced in Mammalian Myocardium by Variation in Perfusate Ionic Composition

This study describes the changes produced in the subcellular morphology of mammalian myocardium when perfusate sodium, calcium, and chloride concentrations are varied. By means of a recently developed perfusion technique, functioning dog papillary muscles were perfused with isotonic solutions of varying ionic compositions. Examination of the tissue in the electron microscope revealed that contr...
